Valaciclovir is rapidly and almost completely converted to aciclovir, then converted to acyclovir monophosphate by a virus-specific enzyme thymidine kinase (TK), then further converted to acyclovir triphosphate by other cellular enzymes. Aciclovir triphosphate competitively inhibits viral DNA synthesis and replication by competing with deoxyguanosine triphosphate for viral DNA polymerase and being incorporated into viral DNA.
